How are antibodies and antigens reacting in the column and what actually causes that positive reaction? Positive reactions are seen by various degrees of agglutination from the top of the column downward. Anti-IgG coating the inside of that gel column will bind to any IgG antibody that’s coating the donor cells, causing agglutination. That agglutination will not allow the red cells to travel down the gel column. If there is no antibody coating the red cells, the anti-IgG doesn’t react with anything, so the red cells freely travel down the gel column indicating a negative reaction.
