🧬 Nobel Prize 2025 in Physiology or Medicine: Part 1 Penelitian oleh Prof. Shimon Sakaguchi yang pada tahun 2025 menerima Nobel Prize in Physiology or Medicine, membuka jalan bagi penemuan Regulatory T cells (Treg). Dalam studi klasiknya (J. Immunol., 1995), limfosit dari tikus BALB/c normal (nu/+) diisolasi dan dibagi menjadi lima kelompok: 1️⃣ Whole lymphocytes (semua sel) 2️⃣ CD25⁻ lymphocytes (semua sel, CD25⁺ dihilangkan) 3️⃣ CD4⁺ CD25⁻ (hanya T helper tanpa CD25) 4️⃣ CD4⁺ CD25⁺ (T helper dengan CD25) 5️⃣ CD8⁺ CD25⁻ (T sitotoksik tanpa CD25) Kelima fraksi ini ditransfer ke tikus BALB/c nu/nu (athymic) yang tidak memiliki timus. Hasil setelah ±3 minggu pengamatan menunjukkan: • Kelompok CD25⁻ dan CD4⁺ CD25⁻ mengalami autoimunitas multipel (gastritis, oophoritis, thyroiditis, glomerulonefritis). • Kelompok whole lymphocytes, CD4⁺ CD25⁺, dan CD8⁺ CD25⁻ tetap sehat. 🧩 Kesimpulan awal: sel CD4⁺ CD25⁺ berperan menekan aktivasi sel T autoreaktif dan menjaga toleransi The work of Shimon Sakaguchi, awarded the 2025 Nobel Prize in Physiology or Medicine, was pivotal in identifying Regulatory T cells (Treg). In his 1995 Journal of Immunology study, Sakaguchi isolated lymphocytes from normal BALB/c (nu/+) mice and divided them into five groups: 1️⃣ Whole lymphocytes – all cells untreated 2️⃣ CD25⁻ lymphocytes – all cells with CD25⁺ removed 3️⃣ CD4⁺CD25⁻ – helper T cells without CD25 4️⃣ CD4⁺CD25⁺ – helper T cells expressing CD25 5️⃣ CD8⁺CD25⁻ – cytotoxic T cells without CD25 Each group was transferred into BALB/c nu/nu (athymic) mice, which lack a thymus. After about 3 weeks, the results were striking: • CD25⁻ and CD4⁺CD25⁻ groups developed multiple autoimmune diseases (gastritis, oophoritis, thyroiditis). • CD4⁺CD25⁺, CD8⁺CD25⁻, and whole lymphocyte groups remained healthy. 🧩 Conclusion: CD4⁺CD25⁺ cells act as regulatory T cells that suppress autoreactive T cells and maintain immune self-tolerance. #physiology #immunology #immunity #nobelprize
