In his 1995 Journal of Immunology study, Sakaguchi isolated lymphocytes from normal BALB/c (nu/+) mice and divided them into five groups: 1️⃣ Whole lymphocytes – all cells untreated 2️⃣ CD25⁻ lymphocytes – all cells with CD25⁺ removed 3️⃣ CD4⁺CD25⁻ – helper T cells without CD25 4️⃣ CD4⁺CD25⁺ – helper T cells expressing CD25 5️⃣ CD8⁺CD25⁻ – cytotoxic T cells without CD25 Each group was transferred into BALB/c nu/nu (athymic) mice, which lack a thymus.
